Effect of Bupeuri Radix on CRF, c-Fos, ACTH and TH in Forced Swimming Test

Abstract
Objectives: The goal of this study was to investigate the antidepressive effect of Bupleuri Radix (BR).

Methods : The forced swimming test (FST) was performed. Also the expression of corticotropin releasing factor (CRF), c-Fos and tyrosine hydroxylase (TH) was measured immunohistochemically at paraventricular nucleus (PVN) and locus coeruleus (LC). Concentration of adrenocorticotrophic hormone (ACTH) was measured in plasma by ELISA method.
Results : The immobility in BR400 Group was significantly decreased in comparison with the control group (p<0.05). The expression of CRF has significantly decreased in BR400 Group (p<0.01). BR100 Group and BR400 Group demonstrated a increase tendency in expression of c-Fos. The expression of TH has significantly decreased in BR400 Group (LC: p<0.001, VTA: p<0.01). In the concentration of ACTH. BR100 Group and BR400 Group demonstrated an increase or decrease, more or less, in comparison with the control group but with no significance.

Conclusions: Judging from the result of the aforementioned tests, Bupleuri Radix had an effect of controlling motionless behavior in FST. Such effect is deemed to have resulted from Bupleuri Radixs controlling CRF in PVN and also TH in LC and VTA. Therefore, it is believed that Bupleuri Radix has an anti-depressant effect of controlling depressive behavior in rats.
